Various 6,7-dihydro-5H-benzo[6,7]cyclohepta[1,2-d][pyrimidines bearing amino acid group at the 4-position of the skeleton were synthesized by the reaction of 4-chloro-6,7-dihydro-5H-benzo[6,7]cyclohepta[1,2-d]-pyrimidine with some amino acid, which were cyclized to 5,6-dihydro-4H-benzo[3,4]cyclohept[1,2-e]imidazo-[1,2-c]pyrimidines, corresponding to B-homo-11,13,15-triazasteroids. Their inhibitory activities against platelet aggregation induced by collagen were also investigated.  相似文献

Reactions of CrCl3(thf)3 with bis(imino)pyridines gave a series of {bis(imino)pyridine}chromium(III) trichloride complexes, {2,6‐(RN?CMe)2C5H3N}CrCl3 [R = C6HPr2‐2,6 ( 1 ), C6H3Et2‐2,6 ( 2 ), C6H3Me2‐2,6 ( 3 ), C6H2Me3‐2,4,6 ( 4 ), C6H3Me2‐3,5 ( 5 ), C6H5 ( 6 ), cyclohexyl ( 7 ), 2‐methyl‐1‐naphthyl ( 8 ), C6H3F2‐2,6 ( 9 ), C6H3Br2‐2,6 ( 10 ), C6F5 ( 11 )]. Pseudo‐octahedral geometries of 6 , 10 , and 11 were revealed by X‐ray crystallography. The complexes having bulky substituents such as 1 – 4 showed high activity for ethylene polymerization in combination with modified methylaluminoxane (MMAO) to give linear polyethylenes. In sharp contrast, the pentafluorophenyl complex 11 /modified methylaluminoxane system was found to be moderately active for ethylene homopolymerization to give moderately branched polyethylene with only ethyl branches. © 2005 Wiley Periodicals, Inc. The interactions of epothilone analogs with the paclitaxel binding site of microtubules were studied. The influence of chemical modifications in the C15 side chain and in C12 on binding affinity and microtubule elongation was characterized. Modifications favorable for binding affinity are (1). a thiomethyl group at C21 of the thiazole side chain, (2). a methyl group at C12 in S configuration, (3). a pyridine side chain with C15 in S configuration, and (4). a cyclopropyl moiety between C12 and C13. The same modification in different ligands has similar effect on affinity, allowing good structure-affinity characterization. The correlation between binding, microtubule stabilization, and cytotoxicity of the compounds has been determined, showing differential effects of the modifications. The binding constants correlate well with IC(50) values, demonstrating that affinity measurements are a useful tool for drug design.  相似文献

Binary sols and gels of SiO2-Al2O3 were prepared using tetraethyl orthosilicate and each of four aluminum compounds; aluminum di (sec-butoxide) ethylacetoacetic ester chelate (AC), aluminum nitrate nonahydrate (AN), aluminum formoacetate (AF), and boehmite sol (BS) made from aluminum i-propoxide. The structure and the evolution of the Si-O-Al bonds in SiO2-Al2O3 sols and gels were investigated by 27Al nuclear magnetic resonance (NMR), Infrared absorption spectra, DTA, and X-ray diffraction. The formation of Si-O-Al bonds differs depending on the aluminum compounds used as raw materials. The ratio of Al(IV) to {Al(IV) + Al(VI)} is related to the microstructural homogeneity of the gels. When AC is used as a raw material, the Si-O-Al bonds are formed in the sol state and resultant gel shows good microstructural homogeneity. In case of AN, the Si-O-Al bonds are not formed either in the sol or the wet gel state. The bonds are formed by drying the gel before heat-treatment temperature reaches 300 400°C, resulting in good microstructural homogeneous gel. When AF is used, the Si-O-Al bonds are formed in the sol state but the ratio of Al(IV) to {Al(IV) + Al(VI)} is lower than when using AC. Microstructural homogeneity of the gel is ranked between AC or AN and BS. Using BS, the Si-O-Al bonds are not formed in the sol solution, and the change in the coordination number of the gel is similar to that of boehmite gel. The Microstructural homogeneity of the gel is the worst among the BS gels, which were prepared by using the four aluminum raw materials.  相似文献

Fragment molecular orbital–interfragment interaction energy calculations of the vitamin D receptor (VDR)/1,25-dihydroxyvitamin D3 complex were utilized to assign functions of key residues of the VDR. Only one residue forms a significant interaction with the corresponding hydroxy group of the ligand, although two residues are located around each hydroxy group. The degradation of binding affinity for derivatives upon removal of a hydroxy group is closely related to the trend in the strength of the hydrogen bonds. Type II hereditary rickets due to an Arg274 point mutation is caused by the lack of the strongest hydrogen bond.  相似文献

Bicyclic 3aH-cyclopentene[8]annulene-1,4-(5H,9aH)-diones underwent three types of acid-induced transannular reactions, Michael cyclization, [3 + 2] cycloaddition, and Friedel-Crafts ipso-alkylation, depending on the cyclopentenone ring substituent (Me or Ph) and the position of [8]annulenone substituent as well as the nature of acids (BF3, MeSO3H, CF3SO3H). The Me-substituent permitted the Michael reaction for all acids used to give tricyclic diones by the activation of cyclopentenone carbonyl group. However, the Ph-substituent inhibited the Michael reaction for BF3 and MeSO3H but allowed the [3 + 2] cycloaddition and Friedel-Crafts reaction for CF3SO3H depending on the position of annulenone substituent. These CF3SO3H reactions exhibited the following novel rearrangements, affording 2-naphthalenone and 7-acenaphthylene derivatives, respectively. The factors that control the reaction mode of these transannular cyclizations were discussed in view of the constraint twist-boat conformation of [8]annulenone ring as well as the ring substituent effects on the intramolecular cyclization. In addition, these [8]annulenone rings were found to easily undergo the intramolecular [2 + 2] photocyclization to provide the tetracyclic cage compounds which exhibited the facile cycloreversion under the influence of acid.  相似文献

 The reaction of 2-amino-4,5-dihydro-3-furancarboxamides with morpholine in the presence of acetic acid in pyridine or under the influence of ammonium acetate gave the corresponding 3-diaminomethylene-4,5-dihydro-2(3H )-furanones; 4,5-dihydro-2-morpholino-3-furancarboxamides were not isolated. One of the former reacted with benzylamine to give (E )- and (Z )-3-(amino-(benzylamino)-methylene)-4,5-dihydro-4-phenyl-2(3H )-furanones and 2-benzylamino-4,5-dihydro-4-phenyl-3-furancarboxamide.  相似文献

The enzymatic cleavage of RNA takes place via a cyclic pentacoordinate oxyphosphorane intermediate/transition state. We carried out ab initio investigations on the neutral cyclic oxyphosphorane, which exists as a stable intermediate. As a consequence of the conformational preferences of the pentacoordinate trigonal bipyramidal intermediates, the rotation of the P-OH bonds is strongly coupled with the reaction coordinate for the pseudorotation process. In addition, the neutral PF(4)OH species has a higher barrier to pseudorotation than the corresponding anionic species PF(4)O(-). These findings are related to the positive charge of the hydrogen atoms on the equatorial oxygens in the trigonal bipyramidal structures: the hydrogen atoms preferably adopt eclipsed positions relative to the axial ligands. Fixing the cationic species in these regions causes an increase in the barrier heights for pseudorotation processes and, thus, prevents isomerization by pseudorotation. Consequently, metal coordination in the double-metal ion mechanism for enzymatic cleavage of RNA should serve to exclusively stabilize the trigonal bipyramidal intermediate/transition state for the in-line attack and departure process.  相似文献

By using an in vivo hydroponic rice seedling culture system, we investigated the physiological and biochemical responses of a model rice japonica cultivar Nipponbare to salt stress using proteomics and classical biochemical methods. Yoshida's nutrient solution (YS) was used to grow rice seedlings. YS-grown 18-day-old seedlings manifested highly stable and reproducible symptoms, prominently the wilting and browning of the 3rd leaf, reduced photosynthetic activity, inhibition in overall seedling growth, and failure to develop new (5th) leaf, when subjected to salt stress by transferring them to YS containing 130 mM NaCl for 4 days. As leaf response to salt stress is least investigated in rice by proteomics, we used the 3rd leaf as source material. A comparison of 2-DE protein profiles between the untreated control and salt-stressed 3rd leaves revealed 55 differentially expressed CBB-stained spots, where 47 spots were increased over the control. Of these changed spots, the identity of 33 protein spots (27 increased and 5 decreased) was determined by nESI-LC-MS/MS. Most of these identified proteins belonged to major metabolic processes like photosynthetic carbon dioxide assimilation and photorespiration, suggesting a good correlation between salt stress-responsive proteins and leaf morphology. Moreover, 2-DE immunoblot and enzymatic activity analyses of 3rd leaves revealed remarkable changes in the key marker enzymes associated with oxidative damage to salt stress: ascorbate peroxidase and lipid peroxidation were induced, and catalase was suppressed. These results demonstrate that hydroponic culture system is best suited for proteomics of salt stress in rice seedling.  相似文献

Volatile pyrolysates of a methyl methacrylate-butadiene-styrene copolymer (MBS) have been analyzed using a capillary gas chromatograph equipped with a Fourier transform infrared detector in tandem with a mass selective detector, and a gas chromatograph-atomic emission detector system. Among the volatile compounds observed were monomers used for synthesis of the polymer. Numerous oligomers of higher boiling point were also found; identification of these could be used to give structural information about the parent polymer. Combining information from these techniques was found to be extremely useful for the analysis of polymer pyrolysis products. In characterizing each compound it was found particularly helpful to juxtapose feature-specific chromatograms (e.g., single ion monitoring from the mass-selective detector and the selected wavelength chromatography from the infrared detector).  相似文献
